Abstract

Maintenance of the proteasome is critical to the survival of all living creatures. Various genes and transcription factors aid in the maintenance of this complex system; amongst them is the protein Heat Shock Factor 1. HSF1 is a highly conserved transcription factor that aids the cell in DNA repair and reparation of other cellular structures in eukaryotes when the organism is put under stress. Caenorhabditis elegans is a model organism that contains homologs to the gene for Heat shock factor 1. It has been observed that C. elegans can survive extended periods of time without food ,and this has been attributed to the HSF pathway. An RNAi feeding vector will be created and fed to the nematodes in order to silence the HSF1 gene. RNAi is a technique that removes an organism’s ability to express a certain gene by targeting messenger RNA for degradation. By removing heat shock factor from the nematodes I hope to confirm that the HSF pathway contributes to their response to stress. It is expected that worms that have undergone the RNAi treatment will not recover from stress placed on them.
